Peptides and the Brain

Interest in “nootropic” and mood-supporting peptides has grown, but the field deserves a careful eye. A handful of peptides can cross into the brain and influence neurotransmitters or neurotrophic signaling — the most studied is Selank, an anxiolytic peptide.

We’ll be clear throughout: these are early-stage compounds. They are not a substitute for evidence-based treatment of anxiety, depression, or other conditions, and anyone struggling should be under the care of a qualified mental-health professional.

Selank: Calm Without Sedation

Selank is a synthetic heptapeptide, a stabilized analog of the immune peptide tuftsin, studied primarily as a non-sedating anxiolytic. Its proposed effects are tied to modulation of the brain’s GABA and monoamine (serotonin) systems — the pathways that govern calm and mood — without the blunt sedation of GABA-receptor drugs.[2]

It has also been reported to raise brain-derived neurotrophic factor (BDNF), a protein central to learning, memory, and neuronal resilience, which is the rationale behind its use for focus and cognitive endurance under stress.[3]

How Strong Is the Evidence?

Most Selank data are preclinical, supplemented by a smaller body of Russian clinical research. One clinical study reported Selank was comparable to a standard benzodiazepine for generalized anxiety, with a favorable tolerability profile and none of the sedation or dependence.[1]

That’s genuinely encouraging — but it’s a limited literature: small trials, largely from a single research tradition, without the large, independent, placebo-controlled replication Western regulators expect.

Our honest framing: Selank is promising but early-stage. It’s used on the basis of mechanism and favorable tolerability signals, not definitive large-scale trials — and it should be weighed against approved anxiety and sleep treatments, not instead of them.
